Protein Production Platform

Do you want to get your hands on a recombinant version of your favourite protein? Have you ever spent months optimising a single protein construct to much frustration?
Established in 2009, the Protein Production Platform (PPP) has systematically cloned, tested and purified multiple protein constructs in tandem, producing more than 4,900 recombinant protein batches for both academic and industry partners. Our strategy is simple, robust and allows us to work in parallel with unique target proteins and multiple protein constructs.
Our Workflow

Your constructs will be sub-cloned and tested for soluble overexpression in Escherichia coli or mammalian cells depending on your needs. We heavily utilize the multiple-construct approach, often employing truncations, solubility tags and alternative expression strains in order to identify well-behaved protein constructs that can be scaled up with ease. Constructs are scored 0 to 4 according to small-scale test results, which highly correlate with scaled-up yield and guide our production decisions.

With proper planning, most targets will produce score-3 and -4 constructs upon first try. However, those with initial scores of 0–2 need not be abandoned. We assess root causes (construct boundaries, tag choice, expression strain) and revise accordingly. Our data show that re-engineering often moves a target from score 1–2 to score 3–4, and the multiple-construct approach facilitates this.
