We are focusing on studying cell stress responses to lipid disequilibrium. Biological membranes contain hundreds of distinct lipids and the composition varies widely among organelles. Therefore, sophisticated homeostasis regulatory mechanisms are needed to maintain lipid homeostasis.
Using a model system reflecting lipid imbalance, we have demonstrated that membrane lipid imbalance activates the membrane stress response (MSR). This response broadly reprograms the protein homeostasis network without changing the lipid composition. Consequently, the MSR buffers the lethal effects of lipid disequilibrium. The unfolded protein response (UPR) arm, a subset of the MSR, maintains membrane functionality without remodeling lipid composition. We are currently studying different aspects of the MSR related to metabolic diseases.
